2",3"-环鸟苷单磷酸-腺苷单磷酸(环GMP-AMP,cGAMP,环[G(2",5")pA(3",5"")p])是在后生动物中发现的第一个环状二核苷酸.2",3"-cGAMP也称为由于在鸟苷和腺苷。2",3"-循环GAMP是一种新的先天免疫第二信使调节I型干扰素(IFN)的产生1-6。由cGAS(cGAMP合成酶)在哺乳动物细胞中产生作为对细胞质中双链DNA与cGAS结合的反应,cGAMP与干扰素基因(STING)。随后STING诱导TBK1-IRF3依赖性IFN-β的产生。

这种cGAS-cGAMP-STING途径已被证明在病原体检测和生理状况,如代谢失调、自身免疫和癌症7-10。cGAMP的细胞浓度由切割磷酸核苷酸的水解酶控制纽带其中一种外核苷酸磷酸酶ENPP1是一种锌刺激的cGAMP水解酶存在于细胞、血清和其他样品中11。

DetectX®2",3"-循环GAMP(cGAMP)免疫测定试剂盒设计用于定量测量存在于裂解的细胞和组织、EDTA血浆和组织培养基样品中的2",3"-cGAMP。在进行此测定之前,请阅读完整的试剂盒说明书。提供2",3"-cGAMP标准生成用于测定的标准曲线,并且应当从标准曲线读取所有样品。清晰提供了用抗体包被以捕获兔IgG的微量滴定板。标准品或稀释样品被移到井中。将2",3"-cGAMP过氧化物酶缀合物添加到标准品和样品中在井里。通过将兔多克隆抗体加入2",3"-cGAMP来引发结合反应每个井。培养2小时后,洗涤板并加入基质。基质反应用结合的2",3"-cGAMP过氧化物酶缀合物,短时间孵育后停止反应并且在能够测量450的微量滴定板读取器中检测产生的颜色的强度nm波长。在制作合适的样品后,计算样品中2",3"-cGAMP的浓度使用大多数读板器可用的软件对样品稀释度进行校正。
